Results of PSI-Blast searches of key TIM barrel sequences


Searching with the sequence of hisA reveals significant homologies to hisF, aroG, fba II, trpC_2, trpA, guaB, pyc, pyrD and other protein families. The PSI-Blast results file is here


KHG/KDPG aldolase (eda) is a Schiff base dependent aldolase it is significantly similar to inosine monophosphate 5'-dehydrogenase (guaB). The PSI-Blast results file is here. Another family of Schiff base dependent enzymes is linked to trpA by KDG aldolase. PSI-Blast file here.


Lactate dehydrogenase (lctD) provides a bridge between dihydroorotate dehydrogenase (pyrD) and pyruvate kinase (pyk). PSI-Blast results here. Pyruvate kinase (pyk), phosphoenolpyruvate synthase (ppsA), and malate synthase (aceB) are linked by citrate lyase (citE). PSI-Blast results file here.


Muconate lactonizing enzyme (MLE) provides a bridge between NIR3 and enolase (eno). PSI-Blast results here. NIR3 found to be significantly similar to dihydroorotate dehydrogenase (pyrD). PSI-Blast results here here. These results also show a significant link to dihyropteroate synthase (folP).


The alpha subunit of tryptophan synthase (trpA) is significantly similar to triose phosphate isomerase (tpi). PSI-Blast results here. trpA is also detected by searches initiated with ribulose 3-phosphate epimerase (rpe), as is thiamin phosphate synthase (thiE). PSI-Blast results here.
